Research statement: Proliferation and differentiation of B-lymphocytes is dependent on receiving a variety of signals from antigens and from interaction with other immune cells. The signals are communicated by cell surface molecules, which induce specific biochemical changes in side B-cells and initiate a program of cell growth and antibody synthesis. We are studying the regulation of such signal transduction mechanisms and their physiological role in the modulation of immune responses, using transgenic mice in which genes for cell surface molecules and the components of the signal transduction pathway are either deleted or overexpressed. One project in the laboratory focuses on a subset of B-lymphocytes that is prone to produce autoantibodies and is elevated in autoimmune states. These concepts are also applied in a system that evaluates the immune responses to bacterial polysaccharides with a goal to increase the efficacy of vaccines against pneumococcal bacteria, which are important pathogens for children and elderly. A third program in the laboratory studies the role of a transcription factor Egr-1 in the growth regulation of normal B cells and B lymphoma cells.
